The redox, structural and spectroscopic properties of native CuA sites are not affected by pH changes at a physiologically relevant range, contrary to what has been observed on engineered CuA sites. This challenges the notion of a previously proposed pH-based mechanism for the regulation of electron transfer reactions of cytochrome c oxidase in vivo.
